Mice that express the human HLA-B27 heavy chain transgene, but with the b2-microglobulin gene deleted, develop arthritis. Free heavy chain has also been detected on human patients with AS, and is shown. HLA-B27 is different from most HLA-B alleles in having a free cysteine at residue 67, and HLA-B27 can form stable dimers without the presence of b2-microglobulin. One hypothetical dimer is depicted. It has been postulated that such a molecule would have a unique motif for the peptides it binds. The presence of such findings radically alters the classical views of pathogenesis. |
