Susceptibility to disease is strongly associated with the MHC class I protein, HLA-B27. Let us further examine the possible role of HLA-B27 in spondyloarthropathy. The MHC, or major histocompatability complex, Class I protein is the star of the presentation and this book. Here its ribbon structure is shown demonstrating the heavy chain's (HC) alpha1, alpha2, alpha3 domains, as well as its transmembrane portion and cytoplasmic tail. The beta2-microglobulin molecule associates with the HC noncovalently. A close-up is made of the MHC groove, as seen by the viewpoint of an approaching T-Cell Receptor (TCR). The alpha helices and the beta-pleated sheets of the alpha1, and alpha2 domains are shown. Peptides which are chemically complementary fit into the groove. A peptide backbone with eight residues is depicted. One characteristic of peptides that fit the HLA B27 motif is that they have an arginine at P2. The arginine anchors the peptide in the B pocket of HLA-B27. A peptide, now containing a space filling molecular model of the side chains of the residues, is shown. When the peptide enters the groove, the P1, P4 and P8 side chains are oriented upward and outward from the MHC groove, and these residues, along with a portion of the proteins comprising the a helices of the MHC, will interact with the TCR. |
